A research consortium for quantitative gene-phenotype relationships in pathogen-attacked barley.
The development of durably pathogen-resistant barley is the main goal of the project. A consortium of phytopathologists and mycologists will address the open questions regarding host susceptability and basal resistance against plant pathogenic fungi from both sides, employing high throughput methods of functional genomics and bioinformatics. The fungi involved are Bipolaris sorokiniana, Rhynchosporium secalis and Blumeria graminis f. sp. hordei.
The group "Phytopathologie und Pflanzenschutz" studies the filamentous, plant pathogenic ascomycete Bipolaris sorokiniana, that causes leaf spot and common root rot on barley and wheat. By random mutagenesis a mutant collection will be generated, that will be tested for disease development on susceptible barley to identify pathogenicity and virulence factors. From all fungi, cDNA libraries are generated and sequenced in high throughput. From the sequence data, interaction microarrays are designed and used to analyse gene expression of the fungi in the course of infection. Candidates for targeted mutagenesis are then identified on basis of the expression data, combined with bioinformatics analyses towards function of the identified genes. The roles of the candidate genes in disease development will be analysed. Mutagenesis will be performed in GFP- and DsRed-tagged strains, that allow microscopic analysis of growth of the mutants in barley leaves because of their green and red fluorescence, respectively. Software tools that are being developed by another member group in the consortium are used to exactly quantify differences in growth between the parental strain and the mutants.
